This presentation will highlight the power of combining total synthesis and late-stage transformations to generate complex, modified proteins for deciphering the molecular roles of post-translational modifications (PTMs) in TFs regulation. Specific examples will focus on the synthesis of site-specifically phosphorylated and acetylated TFs, such as the Myc/Max system.<sup>2, 3</sup> Remarkably, these studies revealed that phosphorylation and acetylation patterns modulate Max–DNA interactions by altering DNA binding affinity and sequence specificity. Importantly, such mechanistic insights led to the development of novel bioactive miniproteins derived from Max (μMax), capable of inhibiting oncogene expression and cancer cell proliferation through antagonistic binding to target genes in cancer cells, paving the way for the development of new therapeutic proteins targeting oncogene expression.<sup>4-6</sup><br>\r\n<br>\r\n<br>\r\n1. It presents around a hundred paintings on wood, didactic works produced between 1997 and 2007 in the teaching units of the painter Arduino Cantàfora. They suggest a possible way of making, between thought and <em>actio</em>, where drawing and painting structure a concept and become an essential language for expressing the founding idea of a project. I then introduce a regularised fracture model, akin to the phase-field regularisation used for softening plasticity, that we recently proposed in [1]. Unlike standard gradient damage or phase-field fracture models, in this approach damage affects the material strength rather than its stiffness.<br>\r\n<br>\r\nThrough analytical and numerical examples, I show how this model, within a consistent variational framework, provides a path toward reconciling several key concepts developed over the centuries, including Griffith and cohesive crack models, damage mechanics, plasticity, strength criteria, and limit analysis.<br>\r\n<br>\r\n[1] B. Target ALS funds investigators who collaborate across scientific disciplines and research constituencies to drive medical breakthroughs for the treatment of ALS.<br>\r\n<br>\r\nAs new therapeutics have begun moving the needle for ALS patients, tracking disease progression and monitoring the effects of drug treatment have become even more important. Biomarkers are needed to stratify patient populations, understand response to therapeutics, and quantify disease progression. The foundation is soliciting proposals from multidisciplinary groups focused on the discovery and development of novel fluid or tissue-based biomarkers of disease progression and stratification using novel, innovative, and complementary approaches including applications of AI and machine learning (ML).<br>\r\n<br>\r\nAlthough all groups studying disease progression biomarkers are eligible to apply, Target ALS is particularly interested in supporting projects that propose the study of:\r\n<ul>\r\n\t<li>novel biomarkers predictive of disease progression or phenoconversion </li>\r\n\t<li>biomarkers with utility as treatment response </li>\r\n\t<li>biomarkers for patient population stratification (e.g., fast vs slow progressors, cognitive impairment, peripheral immunity)</li>\r\n\t<li>novel analytical approaches to blood-based biomarkers, including AI/ML-extracted signatures</li>\r\n</ul>\r\nConfidentiality of the investigator’s data, research, and intellectual property will be strictly honored.  Target ALS does not seek ownership of any intellectual property or financial gains that result from its funding.<br>\r\n<br>\r\n<strong>Funding</strong>:       $130,000 per lab per year (not to exceed a total of $500K / year)<br>\r\n<br>\r\n<strong>Duration</strong>:      24 months <br>\r\n<br>\r\n<strong>Eligibility:</strong> Only <strong>collaborative projects</strong> will be considered; collaborative projects for this call comprise groups of 3-5 laboratories working around a common research theme or target.
