We uncovered a new mechanism for coincidence detection in the Drosophila head direction network. To maintain an accurate sense of direction, head direction neurons that signal orientation during navigation must learn to anchor to relevant external sensory cues in novel environments. Yet the synaptic mechanism for this form of unsupervised learning is unknown in any organism. In Drosophila, GABAergic visual inputs converge onto head direction neurons, and these inhibitory synapses change strength with experience to learn the relationship between visual landmarks and head direction. However, how coincident pre- and postsynaptic activity is detected across this inhibitory synapse is not understood. We discovered that neurons which release the monoamine octopamine close a feedback loop that conveys postsynaptic head direction activity onto presynaptic terminals of visual inputs. This octopamine pathway is required for anchoring the head direction network to visual cues. Furthermore, pairing structured activation of octopamine neurons with a visual cue is sufficient to drive rapid plasticity, even without postsynaptic head direction cell activity. The fundamental unit of chromatin, the nucleosome, consists of ~147 base pairs of DNA wrapped around a histone octamer composed of H2A, H2B, H3, and H4. These nucleosomes are connected by linker DNA, forming a “beads-on-a-string” structure that enables efficient packaging of the genome.<br>\r\nBeyond structural compaction, nucleosomes play a central role in regulating genome function. They can restrict access to DNA and act as barriers to essential processes such as transcription, replication, and repair. At the same time, nucleosomes serve as key epigenetic platforms, where histone modifications and histone variants modulate chromatin structure and dynamics to control gene activity.<br>\r\nTo better understand these regulatory mechanisms, we employ biochemical and structural approaches to investigate nucleosome structure and dynamics. Our studies aim to elucidate how structural transitions of nucleosomes influence DNA accessibility and ultimately govern genomic regulation. I will present our recent structural and biochemical insights and discuss their implications for understanding nucleosome-mediated genome regulation.<br>\r\n<br>\r\n<strong>Bio:</strong><br>\r\nHitoshi Kurumizaka is a Professor at the Institute for Quantitative Biosciences, The University of Tokyo, Japan. After receiving his Ph.D. degree in 1995, he started his postdoctoral training at NIH in the USA, in the laboratory of Molecular Embryology (Alan Wolffe Lab), where he began chromatin research. In 1997, he joined RIKEN as a Research Scientist. He subsequently moved to Waseda University, where he served as Associate Professor and later as Professor of Molecular and Structural Biology. Since 2018, he has been Professor at the Institute for Quantitative Biosciences, The University of Tokyo. His research focuses on the structural basis of chromatin and epigenetic regulation in eukaryotic genomes. In 2020, he established an in-house cryogenic electron microscopy (cryo-EM) platform to advance high-resolution structural studies of chromatin and genome-associated protein complexes. Professor Kurumizaka has made significant contributions to the field of chromatin biology through structural and mechanistic studies of nucleosomes and genome-associated protein complexes. Historically, research has primarily focused on predicting the onset of buckling—that is, the loss of the original configuration under excessive loading—rather than on understanding how structures evolve in the post-buckling regime. Recently, however, the study of buckling has experienced a renaissance, with post-buckling mechanical responses increasingly exploited to achieve new functionalities, for example enhancing fracture resistance in stretchable electronics and flexible displays.<br>\r\n<br>\r\nWhile most existing studies consider elastically deforming systems whose behaviour arises primarily from geometric nonlinearities, this talk presents examples in which geometric effects are coupled with nonlinear constitutive responses, giving rise to new post-buckling mechanisms. First, we investigate the buckling of hard, perforated (“holey”) structures that exhibit auxetic behaviour but, unlike their purely elastic counterparts, undergo post-buckling softening. The resulting negative stiffness is captured using a pseudo-plastic constitutive model that describes nonlinearities emerging from the structural microarchitecture.<br>\r\n<br>\r\nWe then apply a similar framework to examine the buckling of beverage cans largely filled with liquid under axial compression. Whereas empty cylindrical shells typically develop periodic buckling patterns that break both axial and circumferential symmetry, liquid-filled cylinders buckle axisymmetrically. The resulting ring-shaped buckles are localized and emerge sequentially, progressively covering the can surface. We show that this behaviour arises from nonlinear, spatially anisotropic hoop stresses that soften and subsequently re-stiffen during compression.<br>\r\n<br>\r\n<strong>Biography: </strong>Draga Pihler-Puzovic is a Reader in the Department of Physics and Astronomy at the University of Manchester and a core member of the Manchester Centre for Nonlinear Dynamics. Her research focuses on fluid–structure interactions, low-Reynolds-number flows, and the deformation of slender solid structures.
