DESCRIPTION:Prof. Alain Brisson\, Université Bordeaux\nMicroparticles (MP
s)\, also called extracellular vesicles or microvesicles\, are cell membra
ne\nfragments derived from activated or apoptotic cells. MPs attract high
interest as potential disease\nbiomarkers because they contain elements\,
like surface receptors\, specific from their parental cells.\nHowever many
basic questions concerning MPs remain unanswered\, such as: What do MPs f
rom\nblood plasma look like? What is their morphology\, their size distrib
ution? Do they all bind Annexin-5\n(Anx5)? How are they formed? How many a
re they?\nOur overall aim is to answer these questions\, firstly to improv
e our basic understanding on MPs\,\nsecondly to design diagnosis assays on
selected MP sub-populations. Our first objective was to\nprovide a catalo
gue of all types of MPs present in plasma from healthy donors\, considerin
g this as a\nreference for further studies of MPs in various pathological
conditions.\nWe have developed an original approach based on cryo-Transmis
sion Electron Microscopy\n(cryo-EM) imaging and receptor-specific gold lab
eling. Cryo-EM allows revealing the\nvarious types of MPs present in pure
platelet free plasma (PFP). Gold nanoparticles\nfunctionalized with variou
s ligands were developed for identifying specific MP subpopulations.\nWe f
ocused our attention primarily on MPs exposing phosphatidylserine (PS)\,\n
MPs derived from platelets and MPs derived from red blood cells\, using go
 ld particles\nfunctionalized with Anx5\, anti-CD41 and anti-glycophorin an
 tibodies\, respectively
