 ers investigate the dynamic processes of the immune system during the immu
 ne response to pathogens. The main focus is on the function of chemokines 
 and chemokine receptors for the space-time distribution of T-cells\, B-cel
 ls and various kinds of dendritic cells. Processes of cellular differentia
 tion\, activation/suppression\, as well as adhesion/communication\, which 
 are studied in vitro\, but mainly in vivo with the help of mouse models\,

 continuous migration of DC and T cells from blood the periphery into drain
 ing lymph nodes is essential for the induction of protective immunity agai
 nst pathogens as well as for the induction of tolerance towards harmless e
 nvironmental antigens. It is well established that\, following their activ
 ation\, the chemokine receptor CCR7 plays a fundamental role in guiding sk
 in-resident DC towards and into terminal lymphatics. We established recent
 ly intra lymphatic injections of immune cells into lymphatic vessels drain
 ing towards the popliteal lymph node in mice. Using this technique we coul
 d show that DCs enter the draining lymph node by directly penetrating the 
 floor of subcapsular sinus and directionaly migrate into the T cell-rich p
 aracortex. In contrast\, naïve T cells arriving via afferent lymphatics h
 ome into lymph nodes via the medullary sinus. DCs require CCR7 to leave th
 e subcapsular sinus and T cells rely on this receptor for trans-positionin
 g from the medullary sinus into the lymph node parenchyma suggesting that 
 these cells sense a gradient of CCR7-ligands. We now report how such a gra
 dient is built and maintained in the subcapsular sinus of a lymph node.
