DESCRIPTION:Benjamin Rappaz\, McGill University\, Montréal\, Canada\nDuri
ng central nervous system development neurons extend axons over relatively
great distance to reach their ultimate synaptic targets. Netrin is one of
the main axon guidance cues whose effect is mediated by its two receptors
Unc5 and DCC. DCC alone is known to promote attraction\, whereas DCC in c
ombination with Unc5 promotes repulsion.\nMy current project proposes to i
nvestigate the molecular interactions between Netrin and its receptors\, a
nd part of the downstream signaling mechanisms involved using state of the
art optical imaging techniques in living neurons.\nImage Cross-Correlatio
n Spectroscopy (ICCS) and Fluorescence Resonance Energy Transfer (FRET) ar
e two complementary imaging techniques which measure co-localization of fl
uorescently tagged proteins.\nIn this presentation\, I will introduce the
ICCS theory and present Unc5/DCC colocalisation data I obtained with this
technique. FRET was then used to measure the spatio-temporal activation of
Cdc42\, a small Rho GTPase activated downstream of DCC which promotes fil
 ipodia growth.
