 he peroxisome proliferator-activated receptor gamma coactivator 1alpha (PG
 C-1alpha) is a central regulator of fiber plasticity in endurance-trained 
 skeletal muscle. Upon activation by physical activity\, PGC-1alpha control
 s the metabolic and myofibrillar programs that are prototypical of oxidati
 ve\, high-endurance muscle fibers. We now provide evidence that in additio
 n to boosting lipid oxidation\, PGC-1alpha also directly regulates anaboli
 c pathways in muscle\, in particular de novo lipogenesis. Thereby\, storag
 e of lipids is promoted\; these triglycerides are the substrates for ATP g
 eneration during endurance exercise bouts in trained athletes. Our finding
 s have important cautionary implications for the development and applicati
 on of "exercise mimetics"\, drugs that are designed to elicit an endurance
 -trained phenotype even in sedentary individuals.
