DESCRIPTION:Dr. Johannes F. Buyel\, M. Sc.\, Institute for Molecular Biote
chnology\, RWTH Aachen University\nMany feed streams in the biopharmaceuti
cal production of proteins\, e.g. monoclonal antibodies\, have to be clari
fied before the dissolved target can be isolated\, e.g. by chromatography.
Disposable filters\, flocculants and precipitation methods offer a cost-e
ffective\, fast and safe way to achieve this goal. After clarification\, c
hromatographic unit operations are most frequently used to purify the targ
et protein until the required purity is reached\, typically >99% for pharm
aceutical applications. The seminar will highlight some recent achievement
s in clarifying biological feedstocks derived from plants and then focus o
n the rational design and optimization of downstream purification strategi
es for recombinant target proteins. Topics such as modeling chromatographi
c separations and planning experimental verification will be covered.
