DESCRIPTION:Prof. Michael Springer\, Harvard Medical School\, Boston\, MA
(USA)\nBIOENGINEERING SEMINARAbstract:\nThe response of yeast to galactose
is one of the best-studied Eukaryotic signaling pathways. Glucose\, the p
referred carbon source\, inhibits the utilization of galactose. Surprising
ly\, we have found that instead of simply inhibiting galactose utilization
when glucose is above a threshold concentration\, individual cells respon
d to the ratio of glucose and galactose and based on this ratio determine
whether to induce genes involved in galactose metabolism. This response ap
pears to be the result of an extra layer of signal processing which is occ
urring at the hexose transporters.\nFurthermore\, we have uncovered natura
l variation in the threshold of this ratio sensing amongst different natur
al yeast strain. Competitive growth experiments\, nutrient utilization exp
eriments\, and gene induction show that cells display two distinct behavio
rs - a preparation for galactose or an optimization for glucose. These res
ults suggest that the interaction between multiple nutrient sources maybe
more nuanced than previously believed with ratio sensing allowing cells to
behave more optimally to both steady-state and dynamically changing envir
onments.Bio:\nThe Springer lab is interested in how evolution shapes and c
onstrains how organisms respond to their environment. They analyze cellula
r responses in several related yeast species using a combination of in viv
o fluorescence\, synthetic and genetic approaches\, and numerical and anal
 ytical modeling.
