 ally active small molecules remains a great challenge in modern drug disco
 very. Importantly\, rigorous assessment of in vivo target inhibition (targ
 et engagement) has emerged as an important parameter during drug developme
 nt campaigns. In this presentation we demonstrate the power of enzymology 
 combined with metabolomics to identify the exact metabolic target inhibite
 d by –chloro-D-alanine (BCDA)\, a canonical alanine racemase inhibito
 r. Using a combination of enzymology and metabolomics we reveal that BCDA 
 kills Mycobacterium tuberculosis by a completely new mechanism. Stable iso
 tope labelling coupled to metabolomics allow direct assessment of target e
 ngagement by BCDA\, allowing direct monitoring of enzyme and pathway inhib
 ition.
