DESCRIPTION:Ralf Jungmann\, Ph.D.\, Max Planck Institute of Biochemistry\,
  Munich (D)\nBIOENGINEERING SEMINARAbstract:\nSuper-resolution fluorescenc
e microscopy is a powerful tool for biological research\, but obtaining mu
ltiplexed images for a large number of distinct target species in whole ce
lls and beyond remains challenging. Here we use the transient binding of s
hort fluorescently labeled oligonucleotides (DNA-PAINT\, a variation of po
int accumulation for imaging in nanoscale topography) for simple and easy-
 to-implement multiplexed super-resolution imaging that achieves sub-10-nm
spatial resolution in vitro on synthetic DNA structures.\nWe report a mult
iplexing approach (Exchange-PAINT) that allows sequential imaging of multi
ple targets using only a single dye and a single laser source. We experime
ntally demonstrate ten-color super-resolution imaging in vitro on syntheti
c DNA structures as well as four-color two-dimensional imaging and three-c
olor 3D imaging of proteins in fixed cells.\nFinally\, we demonstrate whol
e cell imaging using DNA- and Exchange-PAINT and optical sectioning\, now
allowing DNA-based super-resolution imaging deep inside cells\, away from
the glass coverslip.Bio:\nSince 2014  Independent group leader at the Max
