 a powerful model system to study the early events of tuberculosis. This sy
 stem offers two major advantages: optical clarity and genetic tractability
 . We use real-time microscopy to visualize the sequential stages of the ho
 st-bacterium interaction in experimentally infected animals. We modulate t
 hese stages by creating defined mutations in both the host and the pathoge
 n\, and we determine how these stages contribute to infection outcomes. Ou
 r studies have provided some surprising new insights into the role of the 
 tuberculous granuloma\, the hallmark tissue structure of tuberculosis\, in
  the expansion and dissemination of infection within the host.
