DESCRIPTION:Dr Nicolas Berthet (Institut Pasteur)\nThe development of DNA
microarray as diagnostic tool for identification of pathogens offers inter
esting perspectives. Indeed\, this technology allows unbiased and simultan
eous identification of a huge number of bacterial and viral agents. In thi
s case\, the use of resequencing DNA microarray allows to obtain more prec
ise identification of pathogens and to enlarge the detection spectrum in c
omparison with classical microarray. Until nowadays\, this resequencing mi
croarray was poorly exploited for diagnostic of infectious disease\, parti
cularly in virology. The aim of our work consisted to develop and validate
resequencing microarray for identification of a large series of bacterial
and viral pathogens originating from diverse biological samples. Based on
the Affymetrix technology\, two successive generations of microarrays wer
e developed for detection of both bacterial agents\, associated to their p
athogenicity genes and their conferring antibiotic resistances\, and viral
agents. Our results demonstrated the interest of this technology for path
ogens identification in different clinical situations. Moreover\, discrimi
nation capacity of this microarray was showed for the detection of close v
ariants or for identification of novel variants attached to a viral family
. All together\, our results confirmed advantages of this technology in th
e virology field and open novel routes of optimisation for the development
of a third generation of microarray with enlarged spectrum of viral detec
 tion.
