 \n\n\n	Insulin resistance is associated with the lipid oversupply in obesi
 ty and as such has motivated the search for lipid mediators that can inter
 fere with insulin signaling and glucose homeostasis. One class of lipids e
 levated in tissues of obese humans and rodents include the (glyco)sphingol
 ipids suggesting these bioactive lipids may play a crucial role in insulin
  resistance.  We recently demonstrated that in animal models pharmacologi
 cal reduction of glycosphingolipid biosynthesis ameliorates insulin resist
 ance and prevents some manifestations of metabolic syndrome thereby suppor
 ting this concept. Since the critical (glyco)sphingolipid entities and the
 ir mechanisms are still uncertain\, a systems biology approach is needed t
 o complement classic biochemistry in understanding the role of glycosphing
 olipids in insulin resistance.\n
