 m tuberculosis (Mtb) and nontuberculous mycobacteria (NTM) in culture foll
 owed by the whole genome sequencing of spontaneous resistant mutants has i
 dentified multiple small molecule inhibitors apparently targeting an essen
 tial mycolic acid transporter known as MmpL3. Because of the importance of
  understanding the mechanism of action of these inhibitors to drive their 
 optimization process\, research efforts in our laboratory have focused on 
 characterizing the structure/function of MmpL3\, assessing its therapeutic
  potential in vitro and in vivo\, and developing the whole cell-based and 
 cell-free assays required to assess the mechanism of action and potency of
  MmpL3 inhibitors.
