DESCRIPTION:Prof. Pavol Miskovsky of Safarik University\, Slovakia\nDevelo
pment of selective nano-delivery system and detection of therapeutically a
ctive form of drugs in cell\n\nThe efficacy of photosensitiers (pts) in in
ducing either apoptosis or necrosis after photodynamic action critically d
epends upon different parameters\, including\, in particular: i) selective
pts accumulation in tumor cells and\, ii) dynamics of sub-cellular distri
bution of therapeutically active form of drug.\n\nLow-density lipoproteins
(LDL) play a key role in the delivery of hydrophobic pts to tumor cells i
n PDT\, including hypericin (Hyp). Construction and characterization of ne
w LDL-based selective nano-delivery system is discussed in the presentatio
n.\n\nDynamics of organelle specific sub-cellular redistribution of Hyp pl
ays an important role in its photodynamic activity. In this presentation\,
we report the development and characterization of micrometer-sized dielec
tric beads with nano-metal structures attached to their surface. The metal
ized beads are sufficiently transparent enabling optical trapping while th
e presence of metal nano-islands provided the SERS. This highly efficient
probe can be placed and scanned with nano-metric accuracy in living cells.
Application of such optical nano-sensors is demonstrated on: i) detection
of low quantities of photoactive drug emodine\, (ii) study the kinetics o
f drug diffusion through the cellular membrane towards specific cell organ
 elles.
