 r use of an Anti-Brownian ELectrokinetic (ABEL) trap\, a novel device enab
 ling the study of individual fluorescent nanoscale objects in solution wit
 hout requiring chemical immobilization\, to investigate the conformational
  dynamics of the b2-adrenergic receptor (b2AR)\, a canonical GPCR involved
  in smooth muscle regulation. The ABEL trap uses fast electrokinetic feedb
 ack in a microfluidic configuration to enable direct observation of single
  fluorescently-labelled β2AR molecules for hundreds of milliseconds to se
 conds. By using a conformation-sensitive reporter dye\, we observe a diver
 sity of fluorescence intensity and lifetime levels in individual β2AR mol
 ecules\, indicating changes in radiative lifetime and the existence of a v
 ast range of discrete conformational states. We find that agonist binding 
 increases the dwell times of these states and monitor millisecond fluctuat
 ions within these states.
