DESCRIPTION:Dr. Violeta Castelo Szekely\, CIG\, University of Lausanne\, C
H (until April 2019)\nSEMINAR of the LAUSANNE INTEGRATIVE METABOLISM and N
UTRITION ALLIANCE (LIMNA)\n\nAbstract:\nThe non-canonical initiation facto
r DENR promotes translation reinitiation on mRNAs harbouring upstream open
reading frames (uORFs). Moreover\, DENR depletion shortens circadian peri
od in mouse fibroblasts\, suggesting involvement of uORF usage in clock re
initiation. In this study\, ribosome profiling was used to identify DENR-r
egulated transcripts transcriptome-wide and within the clock circuitry. 24
0 transcripts were uncovered with altered translation rates\, and linear r
egression was used to extract predictive features of DENR dependance. Amon
g core clock genes\, Clock was identified as a DENR target\, whose protein
biosynthesis is regulated through two uORFs and an alternative coding seq
uence start codon. These findings provide insights into uORF-mediated tran
slational regulation that control the mammalian circadian clock and gene e
 xpression at large.\n
