I will describe viral technologies developed by my group that address each of these challenges. These include INTRSECT, a Boolean viral targeting strategy for cell-type- and circuit-specific access; VESPA, a novel split-AAV platform that enables efficient reassembly and expression of genetic payloads too large for a single AAV; and CRISPR/Cas9-based strategies for precise, cell-type- and circuit-specific gene knockdown. Together, these tools form an integrated viral toolkit for programmable access to and manipulation of neural circuits across multiple levels of biological complexity.<br>\r\n<br>\r\n<u><strong>Bio:</strong></u><br>\r\nDr. Lief Fenno is a physician-scientist at The University of Texas at Austin and Dell Medical School. His research integrates systems neuroscience, synthetic biology, and bioengineering to develop genetic, viral, and neuromodulation technologies for programmable control of neural circuits in vivo. His laboratory develops molecular and viral platforms for selectively accessing and manipulating defined neural populations, alongside approaches for neural stimulation and emerging therapeutic applications. His work spans fundamental circuit neuroscience, neurotechnology, and the translation of biological tools to treatment of brain disorders. Corporate debt is not a standard asset with fixed supply and exogenous cash flows. Its market valuation affects debt issuance, which in turn affects aggregate debt supply, default risk, and the cash flows delivered by debt. The model shows how debt demand is an important determinant of leverage dynamics and the cross-sectional distribution of leverage. Importantly, stronger debt demand is entirely absorbed by issuance and leads to lower long-run bond prices in equilibrium. I then extend the framework to allow for imperfect substitutability across bonds. Credit rating segmentation generates leverage dynamics around rating boundaries that are consistent with empirical evidence. When investors have preferred habitats, endogenous demand and supply jointly determine maturity premia. In the first, I'll examine how the presence of co-habiting microbes profoundly affects the biology of a human pathogen, Pseudomonas aeruginosa. This is important, because in diseased human airways - where P. aeruginosa is an unwelcome, but common inhabitant - the pathogen invariably shares the infection site with a variety of other microbes. It turns out that these microbial neighbours interact with one another and that these interactions are perturbed by the loss of a quorum sensing \"master regulator\" (LasR) in P. aeruginosa. In the second \"short\", I'll talk about how we are now able to manipulate the rate of evolution of the organism, either by \"stepping on the gas pedal\" or by \"slamming on the brakes\". The ability to do this, in an experimentally-controlled manner, is allowing us to access some novel evolutionary trajectories, and to address some truly fundamental biological questions. Finally, I'll take a fresh look at bacterial biofilms and in particular, examine whether the exopolysaccharide \"bioglue\" that holds these structures together might have additional - possibly more important roles. It turns out that it does, and that the exopolysaccharide matrix acts as a \"net\" to selectively sequester secreted proteins, potentially conferring novel functionalities on the biofilm.<br>\r\n<br>\r\nBrief Speaker Bio. <br>\r\nAfter reading for a degree in Biochemistry at the University of Oxford, Martin completed a PhD on bacterial chemotaxis at the Weizmann Institute of Science (Israel), where he was awarded the Wolf Prize for his contributions to the field. Leavuing Israel with a PhD and a [German] wife, he then moved to Toulouse (supported by a long-term EMBO fellowship) where he learned the dark art of x-ray crystallography and solved the structure of a signaling complex involved in bacterial chemotaxis. He then moved back to the UK to take up a post-doctoral position at the University of Cambridge working on quorum sensing in the fascinating plant pathogen, Pectobacterium atrosepticum. There, he developed skills in molecular genetics and demonstrated (for the first time) that quorum sensing molecules bind directly to their cognate LuxR homologs in a range of bacteria. This advance allowed him to secure a Royal Society University Research Fellowship to start his own group - where he learned the hard way that prospective students don't like any project with the word \"plant\" in, and when offered projects on the plant pathogen <em>versus </em>a mammalian pathogen, always went for the latter. Since around 2000, his team's focus has been on understanding better the link between metabolism and virulence in the opportunistic human pathogen, <em>Pseudomonas aeruginosa</em>. After receiving her PhD in 1985, de Lange joined Harold Varmus at UCSF for postdoctoral studies. With Varmus, she isolated human telomeric DNA and was the first to show that human telomeres shorten during tumorigenesis. In 1990, she moved to the Rockefeller to take a position as a University Fellow and was appointed as Assistant Professor in 1991. She was promoted to Associate Professor in 1994 and to Professor in 1997. She currently is the Leon Hess Professor and the Director of the Anderson Center for Cancer Research at the Rockefeller University.<br>\r\n<br>\r\nShe is a member of EMBO, the US National Academy of Science, both Dutch Royal Academies, the American Academy of Arts and Sciences, the US National Academy of Medicine. She is an foreign member of the Royal Society of London and received honorary degrees from the University of Utrecht, the University of Chicago, and the University of Groningen.<br>\r\n<br>\r\nDe Lange’s awards include the inaugural Paul Marks Prize for Cancer Research in 2001, the Massachusetts General Hospital Cancer Center Prize, the Judd award from MSKCC, the AACR’s Charlotte Friend and G.H.A.
