In this talk, I will explore scientific autonomy as a process of hill climbing: identifying the capabilities that limit autonomous research, addressing those gaps, and evaluating progress on increasingly demanding chemistry tasks.<br>\r\nDrawing on the El Agente family of systems, I will discuss how hierarchical teams of agents coordinate quantum chemistry workflows and how molecular structure generation addresses a critical gap between chemical reasoning and computational execution. I will consider how these capabilities could contribute to longer-term research goals, such as automated reaction pathway exploration. These examples illustrate how a single missing capability can interrupt an otherwise sophisticated research workflow.<br>\r\nI will also discuss the open challenge of enabling agents to learn continually from scientific literature, computational results, and failed attempts, and to acquire capabilities as new problems demand them. We leverage optogenetics, mechanical perturbations, and modeling to reveal a mechanochemical long-range mutual activation between front and back polarity programs that complements their known local mutual inhibition. Rac-driven protrusions increase membrane tension, triggering mTORC2-dependent Rho activation at the opposite side of the cell. Conversely, Rho-mediated contractility induces cortical-flow regulation of phosphoinositide signaling, promoting distal Rac activation. Our findings demonstrate how the actin cortex and plasma membrane interact as an integrated mechanochemical system for long-range Rac-Rho patterning. Efficient migration also requires polarized organization of the cell interior, with organelle positioning increasingly recognized as a key determinant of cell movement. Yet how this organization is established remains unclear. I will discuss two discoveries that reveal how lysosomes solve these complementary challenges and how their failure creates disease vulnerabilities. First, I will describe Lysosomal Leucine Aminopeptidase (LyLAP), an enzyme specialized to processively degrade the hydrophobic α-helices of transmembrane proteins. LyLAP is highly expressed in pancreatic ductal adenocarcinoma, where its loss causes hydrophobic peptides to accumulate, compromises lysosomal function, and ultimately leads to cancer cell death, revealing a potential vulnerability in tumors with elevated endocytic activity. I will then describe LASER, a damage-responsive protein assembly that couples lysosomal membrane injury to repair. At its core, the oligomeric protein TFG recognizes LC3/GABARAP proteins conjugated to damaged lysosomes and recruits the ESCRT machinery to restore membrane integrity. Neurodegenerative disease-associated mutations in TFG disrupt this process, linking defective lysosomal repair to human disease. The collections thus assembled become true research infrastructures: they provide a unique resource for education and research, accessible to students as well as to innovation communities at EPFL and around the world.<br>\r\n<br>\r\nIt was with this goal in mind that EPFL and the <a href=\"http://verbierfestival.com/\">Verbier Festival</a> launched the Verbier Festival Collection project in 2021. The project aims to preserve and promote the festival’s heritage, with the goal of rekindling younger generations’ interest in classical music.<br>\r\n<br>\r\nA pioneer since 1994, the Verbier Festival is the first festival in the world to have recorded all of its concerts. This exceptional collection now includes 3,000 hours of audio recordings, more than 1,000 hours of video, 12,000 historical photographs, archival documents, and a collection of annotated sheet music. It serves as a unique resource for research, innovation, and cultural transmission.
