Abstract:
Elevated levels of plasma Low-Density-Lipoprotein cholesterol (LDL-C) represent a major risk factor for development of atherosclerosis. Owing to its ability to promote LDL uptake into cells the LDL receptor (LDLR) is an important determinant of plasma LDL level and is a target for human cardiovascular therapy. The LDLR abundance is tightly regulated both on transcriptional and post-transcriptional level, and recently the E3-ubiquitin ligase IDOL was identified as a novel modulator of the LDLR. Acting as an E3 ligase, IDOL supports ubiquitylation of the LDLR on its protruding intracellular tail thereby targeting it for lysosomal degradation, therefore impairing the uptake of LDL in the cell. My talk will focus on the work I carried out on the characterization, regulation and functional implications of this novel regulatory pathway in cholesterol homeostasis.
