We present a strategy based on molecular modelling and simulations for rationalizing and improving Förster resonance energy transfer (FRET) efficiency between donor and acceptor fluorophores in a green fluorescent protein (GFP)-based protein indicator for cyclic adenosine monophosphate (cAMP).
