PGC1 alpha and PGC1 beta are transcriptional coactivators able to upregulate mitochondrial biogenesis, oxidative phosphorylation and fatty acid β-oxidation with the final aim of providing a more efficient pathway for aerobic energy production. In the continuously renewed intestinal epithelium, highly glycolytic proliferative cells in the crypts migrate along the villus axis and differentiate into mature enterocytes shifting their metabolism from glycolysis to oxphos and finally undergoing apoptosis. Using novel transgenic gain and loss of function models we will present an intriguing scenario whereby PGC1α and β are able to directly regulate intestinal epithelial cell fate thus modifying mucosa architecture, enterocyte metabolic activity and cancer susceptibility.
