Bio: Colin Sheppard recently moved to Italy, where he is Senior Scientist at the Italian Institute of Technology. Previously he was at National University of Singapore (Professor of Bioengineering), University of Sydney(Professor of Physics (Physical Optics)), and University of Oxford (University Lecturer in Engineering Science).

Phase contrast techniques in the conventional optical microscope are partially coherent in nature. A few of these methods, in particular differential interference contrast, and differential phase contrast, are introduced, and their optical properties compared. Partially coherent imaging can be simulated based on a phase space model. The Wigner function of the object transmission can be regarded as a measure of local phase gradient, analogous to an instantaneous frequency, thus justifying the physical basis of the Wigner approach.
