NAD is a fundamental biological cofactor that is central to metabolic transformations and to energy homeostasis. Signaling enzymes such as sirtuins, which use NAD as a substrate, are regulated by dynamic fluctuations in NAD and nicotinamide levels in cells, thereby controlling fundamental processes such as stress resistance, DNA repair and metabolic reprogramming. The Sauve laboratory investigates the chemistries of NAD and sirtuins in detail and develops novel chemical tools to manipulate these targets in cells. One eventual aim is to develop new approaches to alter cell and tissue biology for therapeutic benefit.
