Understanding how neurons process and relay information by transducing synaptic inputs into action potentials is a fundamental problem in neuroscience. To improve the performances of the microscope, we have implemented the system for two photon excitation. In this case, control and localization of illumination patterns in the axial direction are achieved by combining holographic illumination with the recently introduced technique of temporal focusing. With this method we show that an axial confinement of about 5 μm is reached independently on the lateral extension of the excitation spot.
