We use mouse genetics to investigate the molecular and cellular mechanisms of tumorigenesis in cancers associated with loss of function of the RB pathway. Our analysis of mouse models has revealed that the retinoblastoma tumor suppressor RB and its family members p107 and p130 prevent cancer initiation in specific populations of adult stem/progenitor cells in the liver, the blood, and the lungs. We have also identified key roles for the Notch and Hedgehog signaling pathways in RB-mutant liver and lung cancers, respectively, thereby identifying novel therapeutic targets for the treatment of cancer patients.
