To obtain thin samples from biological tissue and cells in their flash-frozen native state, cryo-FIB milling is nowadays the most popular method, with impressive successes. The alternative, cryo-ultramicrotomy - also known as CEMOVIS - has been overshadowed because of (i) technical difficulties associated to cryo-section generation and collection - in particular the lack of control over adhesion; and (ii) crevasses and compression artefacts.<br>\r\nI will present recent developments with optimization of the experimental workflow, focusing on section adhesion and their support grids. These result in high throughput data acquisition, with typically several tens to hundreds of cryo-tomograms per sample. Revisiting cutting artefacts shows that crevasses alter but 10 % of the sample volume. Compression is heterogeneous, with scale and material-dependent effects. This is altogether a drawback – with no simple computational correction – and an advantage for projects dedicated to non-(or little)-affected objects such as nucleosomes and chromatin.<br>\r\nAltogether, it is time to revive CEMOVIS as a practical alternative to cryo-FIB milling, offering specific advantages: (i) imaging of large areas of any type of sample, from cells to tissues, organs or even entire organisms – and more generally all kinds of hydrated molecular systems ; (ii) the possibility of serial section approaches ; (iii) no beam damage ; and (iv) a specimen thickness that can be tuned down to 30-50 nm, which may be an advantage for high resolution imaging of small molecular complexes.<br>\r\n<br>\r\n<br>\r\n<br>\r\n<br>\r\n<strong>Biography</strong><br>\r\n<br>\r\nAmélie Leforestier is a CNRS research director at the Solid-State Physics Laboratory (LPS, Paris-Saclay, France). After a PhD in Life Sciences on liquid crystalline self-assembly of DNA, she learned cryo electron microscopy during her post-doctoral studies in the group of Jacques Dubochet in Lausanne. Back in France, she set up a cryo-EM facility for soft matter physics at LPS. She is part of an <a href=\"https://equipes.lps.u-psud.fr/sobio/\">interdisciplinary team</a> dedicated to self-assembly processes at work in living systems at the molecular level. The first part of the talk describes use of the whisker system to study embodied sensing and closed-loop sensorimotor control. On the sensory side, I will present models of whisker mechanics and signal encoding by primary sensory neurons. On the motor side, I will describe models of whisker musculature and emerging insights into the circuits that generate whisker movements. I will then outline key challenges in linking sensing to movement within an integrated closed-loop framework. The second part of the talk will highlight how whiskers interact with moving air. Wood Chair of Tumor Biology and Cancer Research. Prior to joining the UCSF faculty, Dr. McCormick pursued cancer-related work with several Bay Area biotechnology firms and held positions with Cetus Corporation (Director of Molecular Biology, 1981-1990; Vice President of Research, 1990-1991) and Chiron Corporation, where he was Vice President of Research. In 1992 he founded Onyx Pharmaceuticals, a company dedicated to developing new cancer therapies, and served as its Chief Scientific Officer until 1996. At Onyx Pharmaceuticals, he initiated drug discovery efforts that led to the approval of Sorafenib in 2005 for treatment of renal cell cancer, and for liver cancer in 2007, and the approval of ONYX-015 in 2006 in China for treatment of nasopharyngeal cancer. In addition, Dr. McCormick’s group led to the identification of the CDK4 kinase inhibitor, Palbociclib, approved for treating advanced breast cancer. Dr. McCormick's current research interests center on ways of targeting Ras proteins and their regulators, including the NF1 protein neurofibromin.<br>\r\n<br>\r\nDr. McCormick was Director of the Helen Diller Family Comprehensive Cancer Center from 1997 to 2014 and he served as President, 2012-2013, for the American Association for Cancer Research. Since 2013, Dr. McCormick has led the National Cancer Institute’s Ras Initiative at the Frederick National Laboratories for Cancer Research overseeing the national effort to develop therapies against Ras-driven cancers. Recent scientific advances have provided an increased understanding of pathogenic mechanisms underlying FTD and are driving the development of potential disease-modifying therapies. The Alzheimer’s Drug Discovery Foundation (ADDF) and The Association for Frontotemporal Degeneration (AFTD) seek to accelerate this progress by supporting <strong>innovative small molecule and biologic</strong> (antibodies, oligonucleotides, peptides, gene therapy etc.) <strong>drug development programs for FTD</strong> through this request for proposals (RFP).<br>\r\n<br>\r\nThe funding priorities for this RFP are:\r\n<ul>\r\n\t<li><strong>Lead optimization</strong> of novel disease-modifying compounds, including medicinal chemistry refinement and in vitro ADME.</li>\r\n\t<li><strong><em>In vivo</em></strong><strong> testing of novel lead compounds, biologics, vaccines, or repurposed drug candidates</strong> in relevant animal models for pharmacokinetics, dose-range finding, target engagement, in vivo efficacy, and/or preliminary rodent tolerability studies.</li>\r\n</ul>\r\n<br>\r\n<strong>Funding</strong>: up to $300,000 based on stage and scope of research<br>\r\n<br>\r\n<strong>Duration</strong>: 1 year, with potential for follow-up funding<br>\r\n<br>\r\n<strong>Eligibility:</strong> Funding is open to researchers and clinicians worldwide at academic medical centers and universities or nonprofits. Industry partnerships are encouraged.<br>\r\n<br>\r\n<strong>How to Apply</strong>: Applications are reviewed in a two-step process: Letters of Intent (LOI) are the first step, and are submitted through the <a href=\"https://addf.fluxx.io/user_sessions/new\">ADDF Funding Portal</a>. ADDF’s science team will evaluate LOIs to determine whether the proposed project is consistent with the ADDF's mission and funding priorities. LOI decisions will be sent within three weeks of submission. Step 2 is an invited full proposal, which should include a detailed project narrative, budget and justification, and biographical information. When formulating the <a href=\"https://www.alzdiscovery.org/research-and-grants/funding-opportunities/policies\">budget</a>, note that the following costs are not covered: Indirect costs/Overhead, Publication costs, Equipment, and Travel.
