Recent scientific advances have provided an increased understanding of pathogenic mechanisms underlying FTD and are driving the development of potential disease-modifying therapies. The Alzheimer’s Drug Discovery Foundation (ADDF) and The Association for Frontotemporal Degeneration (AFTD) seek to accelerate this progress by supporting <strong>innovative small molecule and biologic</strong> (antibodies, oligonucleotides, peptides, gene therapy etc.) <strong>drug development programs for FTD</strong> through this request for proposals (RFP).<br>\r\n<br>\r\nThe funding priorities for this RFP are:\r\n<ul>\r\n\t<li><strong>Lead optimization</strong> of novel disease-modifying compounds, including medicinal chemistry refinement and in vitro ADME.</li>\r\n\t<li><strong><em>In vivo</em></strong><strong> testing of novel lead compounds, biologics, vaccines, or repurposed drug candidates</strong> in relevant animal models for pharmacokinetics, dose-range finding, target engagement, in vivo efficacy, and/or preliminary rodent tolerability studies.</li>\r\n</ul>\r\n<br>\r\n<strong>Funding</strong>: up to $300,000 based on stage and scope of research<br>\r\n<br>\r\n<strong>Duration</strong>: 1 year, with potential for follow-up funding<br>\r\n<br>\r\n<strong>Eligibility:</strong> Funding is open to researchers and clinicians worldwide at academic medical centers and universities or nonprofits. Industry partnerships are encouraged.<br>\r\n<br>\r\n<strong>How to Apply</strong>: Applications are reviewed in a two-step process: Letters of Intent (LOI) are the first step, and are submitted through the <a href=\"https://addf.fluxx.io/user_sessions/new\">ADDF Funding Portal</a>. ADDF’s science team will evaluate LOIs to determine whether the proposed project is consistent with the ADDF's mission and funding priorities. LOI decisions will be sent within three weeks of submission. Step 2 is an invited full proposal, which should include a detailed project narrative, budget and justification, and biographical information. When formulating the <a href=\"https://www.alzdiscovery.org/research-and-grants/funding-opportunities/policies\">budget</a>, note that the following costs are not covered: Indirect costs/Overhead, Publication costs, Equipment, and Travel. Target ALS funds investigators who collaborate across scientific disciplines and research constituencies to drive medical breakthroughs for the treatment of ALS.<br>\r\n<br>\r\nAs new therapeutics have begun moving the needle for ALS patients, tracking disease progression and monitoring the effects of drug treatment have become even more important. Biomarkers are needed to stratify patient populations, understand response to therapeutics, and quantify disease progression. The foundation is soliciting proposals from multidisciplinary groups focused on the discovery and development of novel fluid or tissue-based biomarkers of disease progression and stratification using novel, innovative, and complementary approaches including applications of AI and machine learning (ML).<br>\r\n<br>\r\nAlthough all groups studying disease progression biomarkers are eligible to apply, Target ALS is particularly interested in supporting projects that propose the study of:\r\n<ul>\r\n\t<li>novel biomarkers predictive of disease progression or phenoconversion </li>\r\n\t<li>biomarkers with utility as treatment response </li>\r\n\t<li>biomarkers for patient population stratification (e.g., fast vs slow progressors, cognitive impairment, peripheral immunity)</li>\r\n\t<li>novel analytical approaches to blood-based biomarkers, including AI/ML-extracted signatures</li>\r\n</ul>\r\nConfidentiality of the investigator’s data, research, and intellectual property will be strictly honored. Target ALS does not seek ownership of any intellectual property or financial gains that result from its funding.<br>\r\n<br>\r\n<strong>Funding</strong>: $130,000 per lab per year (not to exceed a total of $500K / year)<br>\r\n<br>\r\n<strong>Duration</strong>: 24 months <br>\r\n<br>\r\n<strong>Eligibility:</strong> Only <strong>collaborative projects</strong> will be considered; collaborative projects for this call comprise groups of 3-5 laboratories working around a common research theme or target. However, whether this oncogenic process, particularly observed in the intestine, can be initiated by a specific immune cell population remains unknown. We recently identified two distinct effector T cell populations capable to initiate the oncogenic transformation of a healthy intestinal epithelium (Fesneau et al., Nat Immunol. 2024; Brewer et al., Nat. Rev. Canc. 2024), as well as the factors controlling their differentiation.
