To study quantitatively and at a molecular level how the diversity of a population relates to its capacity to respond to selection, we built several minimal antibody libraries and selected them in vitro to bind to specific targets. I will present our analysis of these experiments, which combine antibody phage display with high-throughput sequencing. In particular, I will show how an elementary probabilistic model based on extreme value theory accounts for the diversity of results that we obtain. Our work has several potential implications for designing synthetic protein libraries, for estimating the density of functional biomolecules in sequence space, for characterizing diversity in natural populations and for experimentally investigating the concept of "evolvability ».
