Inflammation initiated by perturbations of organelles integrity

Key cellular functions including those related to energy metabolism, organization of the genetic information or production of membrane-bound and secreted proteins are compartmentalized within organelles. Perturbations of these cellular compartments trigger repair and adaptation programs aimed at restoring homeostasis. These events are often associated with inflammation. Stress pathways stemming from perturbed endoplasmic reticulum (ER) for example have been described as possible contributor to inflammatory responses. Here I will discuss an ER-stress related responses triggered by perturbations of the nuclear envelope (NE). In particular I will focus on the mechanisms linking NE-stress with inflammasome activation.
