The cytosolic receptor RIG-I, which is expressed throughout the body, detects the RNA of invading viruses and is therefore critical in inducing an antiviral immune response, including the production of type I interferons (IFN-I). RIG-I has evolved to recognise viral RNA sensitively but tolerate self-RNA species. Patients with RIG-I gain-of-function mutations often experience a loss of self-tolerance, resulting in the overproduction of IFN-I and the development of autoimmune diseases. In my talk, I will introduce two topics: 'Antiviral innate immune sensing of non-self RNAs' and 'Autoimmune-related innate immune sensing of self RNA'.
