In the last 5 years, the screening of compound libraries against Mycobacterium tuberculosis (Mtb) and nontuberculous mycobacteria (NTM) in culture followed by the whole genome sequencing of spontaneous resistant mutants has identified multiple small molecule inhibitors apparently targeting an essential mycolic acid transporter known as MmpL3. Because of the importance of understanding the mechanism of action of these inhibitors to drive their optimization process, research efforts in our laboratory have focused on characterizing the structure/function of MmpL3, assessing its therapeutic potential in vitro and in vivo, and developing the whole cell-based and cell-free assays required to assess the mechanism of action and potency of MmpL3 inhibitors.
