Classical black-box optimizers struggle in this regime, where evaluation budgets are tight and the feasible region is hard to characterize a priori. Our work investigates how the statistical expressivity of modern generative samplers can be leveraged in black-box optimization. We propose that these samplers enable efficient navigation of feasible sets and optimization of geometrically complex objectives under limited, unreliable observations. <br>\r\n<br>\r\nTwo complementary lines of work have already been studied. The main one, SPARROW, demonstrated that a sampler-driven optimizer can substantially reduce the number of function calls needed to maximize complex and unreliable objectives. A parallel line established that samplers can be steered to satisfy hard constraints without sacrificing the statistical diversity of the generated samples. Because SPARROW imposes minimal assumptions on the underlying sampler, these constrained samplers can drive it directly, opening a path to constrained black-box optimization.<br>\r\n<br>\r\nBuilding on these results, two directions are planned. The first is theoretical: a deeper mathematical study of sampler-driven optimization, drawing on the broader literature on sample-efficient black-box optimization, in order to derive stronger guarantees and more efficient algorithms. With memory accounting for nearly 50% of server costs in modern data centres, provisioning large DRAM capacities significantly increases total cost of ownership (TCO). Tiered memory architectures combine memories with different capacities, latencies, and cost characteristics to improve cost efficiency while satisfying application performance requirements. Application performance in these architectures is highly sensitive to page placement decisions. Consequently, making informed placement decisions requires precise tracking of data access patterns and accurate identification of hot pages.<br>\r\n<br>\r\nExisting operating systems commonly rely on hardware-maintained access bits in page table entries to estimate memory access behaviour and identify hot data regions. They periodically scan the application's entire virtual address space to construct workload access profiles. However, linear scanning of page table access bits does not scale for modern applications with terabyte-scale memory footprints. Since scanning overhead scales with the memory footprint, a single full scan can take several seconds for workloads with large memory footprints. Reducing scanning frequency lowers monitoring overhead but also decreases profiling accuracy and responsiveness to changing access patterns, resulting in a fundamental tradeoff between profiling accuracy and monitoring overhead. Therefore, we need scalable, low-overhead mechanisms for access monitoring in next-generation memory management systems.<br>\r\n<br>\r\nIn this talk, I will discuss recent approaches to scalable memory access profiling in large-memory systems. This problem is exacerbated in state-of-the-art 3D shape generative models that operate in increasingly high-dimensional latent spaces where valid shapes occupy a vanishingly small fraction of the full space. Existing mitigation strategies, including latent regularization and flow-matching approaches, either sacrifice expressiveness, demand a difficult trade-off between objective guidance and generative fidelity that remains prone to manifold drift, or are computationally infeasible to scale to modern, large-capacity 3D shape models. We introduce a novel optimizer-corrector framework that alternates between gradient steps for objective minimization and guided flow matching to drive the latent state back to the valid shape manifold. By decoupling objective minimization from flow-based correction, optimizing freely and correcting strictly, this alternating design avoids inherent trade-offs, preserving geometric validity without sacrificing expressiveness while remaining computationally feasible on modern 3D shape models. Important in industry and medicine, biofilms also represent a unique form of growing active matter with unique physical and material properties. In this talk, I will highlight our recent work on developing a platform for imaging live, growing biofilm at single-cell resolution, using <em>Vibrio cholerae </em>as the model organism. I will show how bacterial cells physically communicate with each other and the environment to form ordered architecture reminiscent of liquid crystals. Moreover, I will talk about our recent discovery that heterogeneity in gene expression leads to unexpected pattern formation in biofilm through a drag-based sorting mechanism. I will also highlight ongoing research in the lab on extending the imaging capability to more natural environments including marine animal hosts. Their atomically thin nature gives rise to distinctive electronic and optical properties, while simultaneously enabling them to function as highly sensitive, readily integrable probes of their local environment. Advanced optical techniques, such as super-resolution microscopy, have opened new opportunities to interrogate these materials at the nanoscale, providing optical access to individual defect behaviors, exciton diffusion and recombination dynamics, and charge transport pathways. Such approaches not only deepen our understanding of intrinsic material behavior but also position 2D systems as powerful sensors capable of resolving local dielectric variations, electric and magnetic fields, and other environmental perturbations.<br>\r\nCentral to this work is hexagonal boron nitride (hBN), a transparent, wide bandgap semiconductor that serves as a host for optically active defects. These defects function as reaction centers, single-photon emitters, and spatially resolved nanoscale sensors that can be readily integrated into electronic and photonic architectures. Owing to its optical transparency, chemical stability, and compatibility with diverse material platforms, hBN provides an exceptional foundation for optical sensing. Building upon this foundation, we explore material properties and present new methods for spatially resolved in situ optical imaging with hBN to advance dynamic, wide-field optical sensing.<br>\r\nWe investigate a previously reported class of emitters that arise from interactions between native hBN and organic solvents. These emitters are believed to originate from defect sites that bind transiently to solvent molecules. To study their behavior, we develop a platform for imaging the dynamics of these transient emitters while varying the electrochemical potential and applying electric fields with controlled orientations. By tracking the spectra of individual emitters, we enable multiplexed measurements that allow spatially resolved electrochemical imaging. Through systematic analysis, we rule out modulation mechanisms based on direct electric field effects or charge transfer. Instead, we identify a mechanism driven by changes in H⁺ concentration, which is modulated during the oxidation of trace water present in the solvent. This finding opens the possibility of using this platform for sensitive detection of H⁺ and trace water in methanol fuel cells, where such species critically influence operational efficiency.<br>\r\nIn the final part of the thesis, we focus on a well-characterized spin defect in hBN, the negatively charged boron vacancy, and explore strategies to enhance its photoluminescence (PL) through heterostructure engineering that facilitates energy and exciton transfer. We demonstrate the coupled structure’s improved utility in optical magnetometry compared to the defect by itself and discuss how improvements in PL could advance the development of wide-field optically detected magnetic resonance (ODMR) imaging using this spin defect. Using a defect in hBN would leverage 2D materials’ exceptional sensing capabilities and planar integrability.<br>\r\nOverall, this thesis aims to highlight the strengths of integrating 2D materials with optical sensing and to develop transferable techniques for introducing controlled stimuli, such as electrochemical potentials, electric fields, or electromagnetic waves, with high fidelity and minimal artifacts.
