Electronic rearrangement is also the means by which light energy is harnessed in photochemistry. The timescale for coherent electron dynamics is set by the energetic splitting of the electronic states, which in small molecular systems, is on the scale of an electron volt (eV). This sets the natural timescale for electronic motion to be few-to-sub femtosecond (fs).<br>\r\n<br>\r\nTo approach these extreme timescales, we can use short pulses of light to excite small quantum systems. For instance, the impulsive interactions between a light field and a quantum system can induce time-dependent oscillations in the charge density. Such electronic wavepacket motion (in the absence of nuclear motion) has come to be referred to as charge migration [1]. While the initial charge dynamics following impulsive excitation (or ionization) begins as purely electronic motion, this wavepacket will couple to other degrees of freedom in the system (i.e. nuclear motion or chemical dynamics) and lead to localization of the charge. The transfer of electronic charge across molecular bonds is fundamental to an understanding of charge transfer phenomena.<br>\r\n<br>\r\nThe study of these fundamental phenomena requires state-of-the-art light sources, such as the Linac Coherent Light Source (LCLS), an X-ray free electron laser (XFEL) facility which produces high-brightness, ultrashort pulses, with wavelength continuously tunable across the x-ray regime. Schemes to provide isolated, sub-femtosecond pulses from an FEL are being explored at facilities world-wide, and recently we have demonstrated such pulses at the LCLS [2]; opening the door for time-resolved measurements of ultrafast electron dynamics on their natural timescale. In my talk, I will highlight our recent developments in probing electronic motion in small molecular systems. We have employed sub-femtosecond pulses from the XFEL to study ultrafast charge dynamics in both core-excited [3,4,5] and low-lying cationic systems [6]. We are also developing nonlinear x-ray spectroscopies [7,8] to initiate and control electron dynamics. To obtain thin samples from biological tissue and cells in their flash-frozen native state, cryo-FIB milling is nowadays the most popular method, with impressive successes. The alternative, cryo-ultramicrotomy - also known as CEMOVIS - has been overshadowed because of (i) technical difficulties associated to cryo-section generation and collection - in particular the lack of control over adhesion; and (ii) crevasses and compression artefacts.<br>\r\nI will present recent developments with optimization of the experimental workflow, focusing on section adhesion and their support grids. These result in high throughput data acquisition, with typically several tens to hundreds of cryo-tomograms per sample. Revisiting cutting artefacts shows that crevasses alter but 10 % of the sample volume. Compression is heterogeneous, with scale and material-dependent effects. This is altogether a drawback – with no simple computational correction – and an advantage for projects dedicated to non-(or little)-affected objects such as nucleosomes and chromatin.<br>\r\nAltogether, it is time to revive CEMOVIS as a practical alternative to cryo-FIB milling, offering specific advantages: (i) imaging of large areas of any type of sample, from cells to tissues, organs or even entire organisms – and more generally all kinds of hydrated molecular systems ; (ii) the possibility of serial section approaches ; (iii) no beam damage ; and (iv) a specimen thickness that can be tuned down to 30-50 nm, which may be an advantage for high resolution imaging of small molecular complexes.<br>\r\n<br>\r\n<br>\r\n<br>\r\n<br>\r\n<strong>Biography</strong><br>\r\n<br>\r\nAmélie Leforestier is a CNRS research director at the Solid-State Physics Laboratory (LPS, Paris-Saclay, France). After a PhD in Life Sciences on liquid crystalline self-assembly of DNA, she learned cryo electron microscopy during her post-doctoral studies in the group of Jacques Dubochet in Lausanne. Back in France, she set up a cryo-EM facility for soft matter physics at LPS. She is part of an <a href=\"https://equipes.lps.u-psud.fr/sobio/\">interdisciplinary team</a> dedicated to self-assembly processes at work in living systems at the molecular level. Its 10th edition, organized in collaboration with the Institute of Architecture, will be entitled <em>Autonomy and Abstraction </em>and will deal two fundamental dimensions of scholarly inquiry. The two days event, open to the public, will be structured in the form of thematic sessions alternating with keynotes and round tables.<br>\r\n<br>\r\n<em>Abstraction</em> will be approached in its classical philosophical meaning, consistent with the view that understanding requires the mind to grasp the universal apart from the particulars in which it is embodied. In this sense, abstraction is the mental operation through which the essential structure of a subject is drawn out from its contingent features, allowing researchers to discern the general principles that render their object intelligible. This conceptual withdrawal does not negate the concrete but clarifies what endures beyond the variability of specific instances.<br>\r\n<br>\r\n<em>Autonomy</em> will be presented as a distinct yet complementary notion: the capacity of a discipline to reflect upon itself through its own internal laws and modes of reasoning. It entails a temporary suspension of external frameworks so that the discipline may be understood from within, according to the coherence of its concepts and the logic that shapes its questions. While abstraction deals with the apprehension of universal principles, autonomy has to do with the conditions under which these principles operate within a disciplinary horizon.<br>\r\n<br>\r\nOnce defined as separate notions, abstraction and autonomy will be brought into constructive relation, for both require a measured intellectual distance that enables the essential to be articulated. In this context, participants will be invited to suspend, for a moment, the temporal and spatial dimensions that ordinarily situate their work, in order to concentrate on the internal logics that define it. <em>The possibility of abstraction and autonomy</em> will be explored not as a doctrine but as an open interpretative field through which doctoral researchers may examine their subjects at the level of foundational assumptions and operative laws.<br>\r\n<br>\r\nThese <em>Rencontres</em> will emphasise that both abstraction and autonomy admit multiple interpretations, each opening further avenues for reflection. Their plurality will form the ground of the collective discussion.
