Membrane insertion constrains both the sequences of transmembrane segments and their placement within proteins. These constraints can conflict with the sequence requirements of protein function. Combining sequence analysis with cellular experiments, I investigate two challenges: inserting C-terminal segments and accommodating hydrophilic segments within the membrane. I will bring together published and ongoing work to explore how insertion pathways shape membrane protein sequences and architecture.
