I will describe viral technologies developed by my group that address each of these challenges. These include INTRSECT, a Boolean viral targeting strategy for cell-type- and circuit-specific access; VESPA, a novel split-AAV platform that enables efficient reassembly and expression of genetic payloads too large for a single AAV; and CRISPR/Cas9-based strategies for precise, cell-type- and circuit-specific gene knockdown. Together, these tools form an integrated viral toolkit for programmable access to and manipulation of neural circuits across multiple levels of biological complexity.<br>\r\n<br>\r\n<u><strong>Bio:</strong></u><br>\r\nDr. Lief Fenno is a physician-scientist at The University of Texas at Austin and Dell Medical School. His research integrates systems neuroscience, synthetic biology, and bioengineering to develop genetic, viral, and neuromodulation technologies for programmable control of neural circuits in vivo. His laboratory develops molecular and viral platforms for selectively accessing and manipulating defined neural populations, alongside approaches for neural stimulation and emerging therapeutic applications. His work spans fundamental circuit neuroscience, neurotechnology, and the translation of biological tools to treatment of brain disorders. In the mammalian clock, PERIOD (PER) proteins form complexes with CRYPTOCHROME (CRY) and casein kinase 1δ (CK1δ) and undergo extensive phosphorylation within intrinsically disordered regions over the circadian cycle.<br>\r\nWe identify a molecular principle by which progressive phosphorylation can generate distinct and persistent regulatory states. Whereas individual phosphosites are rapidly dephosphorylated, multisite phosphoclusters within disordered regions of PER2 become resistant to phosphatases and display switch-like, all-or-none phosphorylation. Distinct phosphoclusters, including the FASP region, the β-TrCP phosphodegron, and a C-terminal phosphocluster, exhibit markedly different dephosphorylation kinetics.<br>\r\nTogether, these findings suggest that progressive phosphorylation of intrinsically disordered PER2 is decoded by phosphoclusters that function as molecular switches with distinct kinetic properties. Their sequential accumulation may generate a series of metastable functional states, thereby translating progressive phosphorylation into circadian control of subunit remodeling, subcellular localization, and turnover of repressor complexes.<br>\r\n<br>\r\n<u><strong>Bio:</strong></u><br>\r\nMichael Brunner is Professor of Biochemistry at Heidelberg University and a leading researcher in the molecular mechanisms of circadian clocks. He received his PhD from Heidelberg University in 1989, followed by postdoctoral research with James E. Rothman at Princeton University and the Sloan Kettering Institute, and with Walter Neupert at Ludwig Maximilian University of Munich. During this time, he made important contributions to mitochondrial biology, particularly to elucidating the molecular machinery responsible for mitochondrial protein import. He joined the Heidelberg University Biochemistry Center (BZH) as a professor in 2000 and has served several terms in its academic leadership, including as Director of the BZH. He is currently Editor-in-Chief of FEBS Letters.<br>\r\n<br>\r\nBrunner’s research has contributed to one of the most complete molecular descriptions of biological timekeeping in any organism. Using the filamentous fungus Neurospora crassa, his laboratory has elucidated central features of the circadian feedback loop, including the functions of the White Collar Complex and FREQUENCY, and the roles of casein kinase 1, progressive phosphorylation, and dynamic complex remodelling in determining circadian period and timing. In parallel, his group has extended this work to human cells, investigating the coordination of the circadian clock with the cell cycle and cellular proliferation. Notably, this research has revealed mechanisms through which oncogenic pathways such as MYC can suppress the molecular clock and promote proliferation, thereby linking circadian regulation to malignant growth. In the study of the cell nucleus, SMLM has offered unique insights into its organization and function at the nanoscale. Nuclear architecture is structured across multiple spatial scales in three dimensions and is highly dynamic. Understanding the mechanisms that govern nuclear organization therefore requires imaging approaches capable of capturing molecular dynamics across a broad range of spatial and temporal scales and in diverse biological contexts. Although SMLM can routinely achieve spatial resolutions in the tens-of-nanometers range, many biologically relevant structures and processes remain beyond its reach. For example, resolving the precise organization of DNA at the molecular scale remains challenging, as it would require a substantial improvement in spatial resolution over conventional SMLM, while also addressing the high density and degree of molecular compaction within the nucleus. <br>\r\nOver the years, we have developed complementary strategies to address these challenges by introducing additional dimensions and observables into SMLM measurements. In this presentation, I will first discuss our approaches to three-dimensional single-molecule imaging, including multifocus microscopy and volumetric light-sheet excitation, which enable fast and sensitive imaging over extended volumes and in complex biological samples such as cell colonies. While these approaches provide access to the three-dimensional and temporal dimensions required to investigate nuclear organization, spatial resolution alone is not always sufficient to distinguish molecular arrangements. We have therefore extended SMLM further by implementing polarization-resolved multifocus microscopy (PolMFM), which combines three-dimensional localization with information on molecular orientation.<br>\r\nI will illustrate how these multidimensional approaches can provide new insights into nuclear organization, with examples spanning different biological systems. In particular, I will present evidence for a long-range, crystal-like organization of DNA within the nuclei of mature sperm cells from crickets, demonstrating how multidimensional imaging can reveal structural features that remain inaccessible to conventional localization microscopy.<br>\r\nFinally, I will discuss the challenges and opportunities associated with visualizing, analyzing, and interactively exploring the multidimensional datasets generated by these approaches. Together, these developments illustrate how extending SMLM beyond conventional two-dimensional localization can provide new ways to interrogate the organization and dynamics of biological systems at the nanoscale.<br>\r\n<br>\r\n<br>\r\n<u><strong>Bio:</strong></u><br>\r\nBassam Hajj is a physicist and CNRS researcher at Institut Curie in Paris, where he is a member of the Physics of Cells and Cancer unit and co-leads the “Light-based Observation and Control of Cellular Organization” team. His research focuses on the development of advanced optical microscopy techniques, particularly single-molecule localization microscopy, 3D imaging, and multidimensional approaches for studying cellular organization at the nanoscale.<br>\r\nAfter completing his PhD at École Normale Supérieure de Cachan on electro-optical microscopy, Bassam joined Janelia Research Campus as a postdoctoral researcher, where he worked on multifocus microscopy for fast and sensitive single-molecule imaging. While safety is paramount, guarantees on sufficient exploration are also crucial for ensuring autonomous task completion. To address these challenges, we propose a novel safe guaranteed exploration framework using optimal control, which achieves first-of-its-kind results: guaranteed exploration for non-linear systems with finite time sample complexity bounds, while being provably safe with arbitrarily high probability. The framework is general and applicable to many real-world scenarios with complex non-linear dynamics and unknown domains. For efficient implementation, we exploit goal-directed exploration, and receding-horizon replanning while preserving the framework’s guarantees, and demonstrate safe, efficient exploration in challenging unknown environments using a car model.<br>\r\n<br>\r\n<strong>Biography</strong>: <br>\r\nManish Prajapat earned his Ph.D. in reinforcement learning and control from ETH Zurich in May 2026. He was a Doctoral Fellow at the ETH AI Center working with Prof. Melanie Zeilinger and Prof. Andreas Krause. Previously, he earned his master’s degree in Robotics, Systems, and Control from ETH Zurich and was a visiting scholar at Caltech. The talk positions older adults as active contributors to robotics research, whose lived expertise can shape both future robot companions and the broader systems of support surrounding them.<br>\r\n<br>\r\n<br>\r\n<strong>Biography</strong><br>\r\nChorong Park, Ph.D., is an Assistant Professor and Presidential Frontier Faculty at the University of Houston’s Gerald D. Hines College of Architecture and Design, where she leads the Empathetic Lifespan AI & Robotics for Aging (ELARA) Lab. Her research connects human–robot interaction, human-centered AI, and participatory design to develop social robots and supportive technologies with older adults and caregivers.<br>\r\n<br>\r\nDrawing on studies in rural Indiana and urban Houston, she investigates how robots’ physical forms and interactions can fit older adults’ needs, surroundings, and everyday lives. Her work centers older adults’ perspectives on companionship, support, privacy, personal boundaries, and dignity, including how caregiver involvement can preserve individual choice and control.<br>\r\n<br>\r\nHer research is informed by more than 500 hours of volunteer technology support with older adults and people with disabilities and has appeared at ACM CHI, ACM ASSETS, and IEEE robotics venues. We leverage optogenetics, mechanical perturbations, and modeling to reveal a mechanochemical long-range mutual activation between front and back polarity programs that complements their known local mutual inhibition. Rac-driven protrusions increase membrane tension, triggering mTORC2-dependent Rho activation at the opposite side of the cell. Conversely, Rho-mediated contractility induces cortical-flow regulation of phosphoinositide signaling, promoting distal Rac activation. Our findings demonstrate how the actin cortex and plasma membrane interact as an integrated mechanochemical system for long-range Rac-Rho patterning. Efficient migration also requires polarized organization of the cell interior, with organelle positioning increasingly recognized as a key determinant of cell movement. Yet how this organization is established remains unclear. I will discuss two discoveries that reveal how lysosomes solve these complementary challenges and how their failure creates disease vulnerabilities. First, I will describe Lysosomal Leucine Aminopeptidase (LyLAP), an enzyme specialized to processively degrade the hydrophobic α-helices of transmembrane proteins. LyLAP is highly expressed in pancreatic ductal adenocarcinoma, where its loss causes hydrophobic peptides to accumulate, compromises lysosomal function, and ultimately leads to cancer cell death, revealing a potential vulnerability in tumors with elevated endocytic activity. I will then describe LASER, a damage-responsive protein assembly that couples lysosomal membrane injury to repair. At its core, the oligomeric protein TFG recognizes LC3/GABARAP proteins conjugated to damaged lysosomes and recruits the ESCRT machinery to restore membrane integrity. Neurodegenerative disease-associated mutations in TFG disrupt this process, linking defective lysosomal repair to human disease.
